In the laboratory, it is often necessary to use cryovials to aliquot cells and tissues into low-temperature storage, but when we take out samples that have been stored for a long time from ultra-low temperature freezers or liquid nitrogen tanks, we are often frightened by the crackling sound of cryovials and cardiac arrest. The bursting of the cryovials will not only cause the loss of experimental samples, but may also cause injury to the experimenter. Therefore, it is important to understand the cause of the tube burst and how to avoid it.

Causes of cryovialized tube bursts

Many times, cells need to be frozen for long-term storage in the experiment, in case of emergency. Long-term storage temperature is -130 °C or lower. The gaseous layer temperature in the liquid nitrogen tank is between -140 °C and -180 °C, and cells can be stored in the gas layer or immersed in liquid nitrogen.

 

The root cause of the bursting of the cryovials is that liquid nitrogen remains in the vials. When the cryopreservation tube is taken out of the liquid nitrogen tank, the temperature in the tube rises, the liquid nitrogen in the tube rapidly vaporizes, from liquid to gas, theoretically 1 volume of liquid nitrogen, in the standard state will vaporize into 646.4 volume of nitrogen. At this time, the frozen storage tube can not remove the excess nitrogen in time, and the nitrogen pressure accumulated in the frozen storage tube increases sharply, and when the tube body cannot withstand the high pressure generated inside, it ruptures, resulting in a burst.

Differences between internally and externally rotated cryovials

Usually we can choose a well-sealed internal rotation cryopreservation tube. From the structure of the tube cover and the tube body, the liquid nitrogen in the internal rotation cryopreservation tube is easier to discharge than the external rotation cryopreservation tube when vaporized, and the design difference of the same quality of the cryopreservation tube will make the sealing of the internal rotation cryopreservation tube better than the external rotation tube, so it is not easy to cause a burst.

However, the bottom of the screw cap has easy access to the sample inside the tube, which may cause sample contamination, and the external screw cap can reduce the chance of contamination when handling the sample. Moreover, the internal volume of the internal rotation cryovials of the same specification will be smaller than that of the external rotation tube.

Operations that reduce the probability of squibing

If there is only an externally rotated cryovial, what methods can reduce the probability of a tube burst?

 

First of all, after we take it out of the liquid nitrogen tank, we need to place the cryovials horizontally or upside down on the table for tens of seconds as soon as possible, and then open the cryovials for the operation of resuscitating cells after the liquid nitrogen flows out.

 

When cryopreservation of cells, you can add more cryopreservation solution. For example, add 1.8ml of cryopreservation solution to a 2ml cryopreservation tube to retain less gas space. Also, when cryopreservation cells, the mouth of the cryopreservation tube should be tightened as much as possible.

 

The use of cryopreservation tube sleeve can isolate the cryovials and liquid nitrogen to a certain extent, and avoid liquid nitrogen directly contacting the cryopreservation tube and penetrating into the tube. Keeping the cryovials in the liquid nitrogen gaseous layer can effectively prevent liquid nitrogen from entering the cryovials. However, only a small amount of liquid nitrogen can be stored in the liquid nitrogen tank, and liquid nitrogen needs to be added frequently.
