Introduction of cryopreservation tubes: cryopreservation tubes are generally divided into 0.5ml, 1.0ml, 1.5ml, 1.8ml, 2.0ml, 4ml, 5ml, 7ml, 10ml, etc. according to their capacity. General cryotubes cannot be put into liquid nitrogen, only those processed with special materials can be put in. At the same time, cryopreservation tubes also have double-layer and no double-layer silicone pads and those without silicone pads, as well as colorless and variegated colors, and various solid colors. If you need to enter the liquid nitrogen storage, you must choose a sealed, low-temperature-resistant special cryotube, and you must also recognize whether the cryotube you buy is sterile and free of DNA and RNA.
How to use the cryotube:
⑴ Pick a fresh culture from the pure culture of bacteria to form a bacterial suspension with a turbidity of about 3-4 McDonnell’s turbidity and inoculate it in a bacterial preservation tube.
⑵ Tighten the preservation tube, turn it upside down 4-5 times to emulsify the bacteria, and do not shake it.
⑶ Put the preservation tube in the refrigerator (-20℃--70℃)
Frequently asked questions about cryopreservation tubes:
1. Question: Can the Nunc cryopreservation tube be sterilized?
Answer: Although the cryopreservation tube is made of polyethylene material, we do not recommend customers to sterilize it.
2. Question: What is the difference between the inner screw cap and the outer screw cap of the cryopreservation tube?
Answer: The inner rotary cryotube is designed for cryopreserving samples in the gas phase, liquid nitrogen phase, and the nozzle silicone pad enhances the tightness of the cryopreservation tube; the outer rotary cryotube is designed for freezing samples in mechanical equipment such as refrigerators. The screw-on screw cap can reduce the chance of contamination when handling samples.
3. Question: Can the Nunc cryopreservation tube be immersed in liquid nitrogen (liquid phase) to freeze samples?
Answer: Nunc's internal rotation cryotubes can be stored in liquid nitrogen, due to the enhanced sealability of the silicone gasket. However, at ultra-low temperatures, any sealing material becomes extremely fragile. In theory, as long as it is a mechanical seal, there is a risk of leakage. In order to prevent the risk of leaking products, the mechanically sealed cryotube must be sealed in a protective bag. Nunc provides CryoFlex sealing tube, heat-sealed polyethylene tube is specially designed for Nunc cryopreservation tube sealing. It is very dangerous to freeze cryopreservation tubes in liquid nitrogen liquid phase without using CryoFlex sealed tubes. CryoFlex sealed tubes must be used to heat seal cryopreservation tubes in liquid nitrogen liquid phase.
4. Question: When the cryotube is stored in liquid nitrogen without using CryoFlex Tubing, how to dissolve and recover cells correctly?
Answer: Protective clothing must be worn during the entire operation, including gloves, face shield, etc. The cryopreservation tube must be stored in the liquid nitrogen phase for 24 hours after being taken out of the liquid nitrogen phase, so that the liquid nitrogen can slowly evaporate and escape from the cryopreservation tube, thereby reducing the pressure in the cryopreservation tube. After 24 hours of storage in the liquid nitrogen phase, put the cryopreservation tube into the water bath while covering it with a towel, and transfer the frozen material after the melting and recovery is complete.
